The sequencing chemistry:  
ABI PRISM BigDye™ or Amersham Pharmacia Biotech DYEnamic ET Terminator chemistry will be used for the sequencing reaction. The BigDye™ or DYEnamic ET Terminator kits are ready for use with any unlabeled primer and purified DNA template. Unincorporated dye terminators will be removed from the sequencing reactions by purification through Qiagen DyeExTM Spin columns.

 

The services:

You perform the sequencing reactions using ABI PRISM BigDye™ or DYEnamic ET Terminator, according to manufacturers’ instructions, and provide us with a dried pellet of purified reaction products ready to run on the sequencer. For optimum results, it is essential that the primers used for PCR are removed from PCR products (by gel or column purification) and that unincorporated dye terminators are removed from the sequencing reaction mix (by ethanol precipitation or purification through QIAGEN DyeEx™ Spin columns).

You provide us with your plasmid DNA (QIAGEN mini- or maxi-prep DNA preferred) or purified PCR product and custom primer (click here for the free universal primers provided). We will verify the quantity and quality of the templates and primers after transportation and perform the sequencing reactions accordingly.  We will then remove excess dye terminators and run the products on a sequencer. These times may vary depending on sample load. Sequences will be e-mailed and electropherograms delivered to you the next day.

You provide us with your template DNA, primers (including information on the Tm of primers) and PCR conditions and we will perform the PCR reactions with QIAGEN Taq polymerase.

You provide us with your PCR product and we will purify your PCR product using the QIAquickTM Gel Extraction Kit. The purified PCR product is ready for sequencing reaction.

You provide us with your template DNA (at least 25 mg QIAGEN midi- or maxi-prep DNA preferred) and we will sequence one strand of the entire insert. Sequencing will be carried out using BigDye™ or DYEnamic ET Terminator chemistry and purified with the QIAGEN DyeEx™  Spin kit. You will receive the completed sequence plus all primers synthesised for the project. Please provide us with as much information as possible about the plasmid/insert (plasmid name, size, restriction sites used to clone the insert, priming sites, any sequence information you currently have, etc).

Time required: Approximately 1 kb/week. Sequences will be e-mailed to you as they are generated, and the completed sequence and electropherogram will be delivered the day after completion.
